Mixed Mode Cation Chromatography Resin


"Hard gel" mixed-mode cation chromatography resin MMC uses polymethacrylate as the matrix. Different from traditional agarose-based resin, "hard gel" MMC exhibits stronger pressure resistance and is particularly suitable for processing high viscosity and high turbidity samples. Through flexible hydrophilic technology, it not only retains the rigid characteristics of the polymer, but also has the good biocompatibility of agarose. In addition, we have launched a series of ultra-large pore (GP) products on this basis, specifically for the purification of macromolecules >100 kD or other ultra-large molecules.

Capacity

 

The unique ligands of "hard gel" MMC not only cover conventional weak cationic functional groups, but also incorporate hydrophobic groups with benzene ring structures. Due to the combination of ion exchange and hydrophobic interaction, the MMC mixed mode weak cation exchange resin can maintain an ideal dynamic loading capacity even in a high-salt environment (see Figure 2).

Applications

 

"Hard gel" MMC can effectively remove aggregates, nucleic acids, host proteins, viruses, and is widely used in purification in various fields, including the purification of recombinant subunit proteins, vaccines (including human vaccines and veterinary vaccines), collagen, and others.


Different target molecules correspond to different application scenarios, and a variety of MMC types are available. When the viscosity of the feed solution is high, it is recommended to choose a polymer matrix. See Figure 3 for detailed selection guide.

The recombinant protein sample in the case in Figure 4 has high viscosity and high conductivity (>20 mS/cm). Conventional ion exchange chromatography requires dilution and diafiltration. However, MMC mixed mode weak cation exchange resin was used later to conduct pH and conductivity-based elution. The experimental results showed that impurities such as nucleic acid and HCP flowed through, and the purity of the eluted sample was >90%.
